Characterization of the Activity and Regulation of Mus81-Mms4/EME1 in Homologous Recombination

Mus81-Mms4/EME1 is a structure-selective endonuclease involved in homologous recombination mediated replication fork support. The activity of this Saccharomyces cerevisiae Mus81-Mms4 has been shown to be regulated by phosphorylation by cell cycle kinases and biochemical approaches will be implemented to elucidate the mechanism of activation. In addition, Homo sapiens MUS81-EME1 has been identified as a potential anti-cancer target. A high throughput inhibitor screen will be conducted to identify candidate MUS81-EME1 inhibitors that may be used as anti-cancer therapeutics.
